February - Finishing and writing

Finishing a piece of science involves reading, final experiments and the condensantion of ideas in short sentences that can be interpreted by a vast audience. It is not an easy goal when something new arises, but connections to what was done before and insight into what can be expected from the knowledge added. February was a month of writing and thinking and a bit of experimentation. Football started, Loschwitzer SV is a new brotherhood of german spielzeugs that have two buttons, pass and run. It reminds me the early british kick and rush system, with ball to the ground and lots of physical power. And this time I play in the middle of the field, where al the heat is on, and you have to know exactly what you are doing because after 3 seconds, already two guys have cut your passing lines and are eagerly prone to take the ball aswell. Discipline and tactics. And the snow is gone. And i just woke up. :)

This post is to honour our Toni, the Dr Toni, 70 year old fussball player that scored an amazing goal today. It was like the one in the video, from the virtual Luca Toni from Bayern in fifa 2009. It was not Bestchastnyk this time, that 12 years ago was destroying defenders in FIFA 1996 in Oeiras, but Luca, the Toni striker from Bayern. Our Luca Toni just chipped the ball on top of a not so cautious keeper, with one beautiful touch of magic. Hail the elder king of football, our TONI!!! :)

When I saw a movie, where death was kept closed inside a bottle, and the most medieval battles did not allow the suffering to perish, I was faced with a dilemma: Is death good or bad?

Independently from that, animals are open systems. They are able to regenerate. So why not live healthier for longer? Without cancer, hearth and brain failures or degenerative diseases.

Blasco, Nystrom, Hayflick, Guarente, Barker and Walsmley, Ackermann, Minois, Taddei, Manuel Serrano and Yves Barral. Your work has inspired me. Mortimer and Johnston, 50 years ago started something amazing. They saw that unicellular organisms died. I saw otherwise.

Coming back to my childhood: Once I woke up crying, telling my father I did not want him to die, or any other person I loved to die. I did not want dinossaurs to be extinguished. I do not want my grandmother to stop cooking, or my mother stop loving me. I do not wish that people I love disappear. But there is one thing... Their happiness depends on my happiness, I cativate people with an unusual easeness, so I am responsible for them. I cannot go crazy looking for the fountain of youth, fighting all the time, I have to respect myself. So I can retribute the love I was given.

So I am not God. That was a shock. Still, I can try to discover how aging works and how Darren Arronovski envisioned the fountain, how Duncan McLaud want to chop his head off, how queen sang want to live for ever! I will understand why to sacrifice a land to gain 2 life was important, without sacrificing myself (zuran orb). I will continue working on aging, because I had a glimpse of immortality in one of my experiments.

Calmly, in an organized fashion, I will look at wath is happening inside the nucleus. Fission yeast is the future. The pandora box has been opened. Aging science will rise, with the new Max-Planck in Cologne.

Fission yeast is immortal. I believe. I cannot prove. But I feel it inside myself. Here lies the secret of immortality, symmetry and repair.

Aging studies continues

We lost 6-2 against brasil, but they never found an immortal organism. I did, and I will prove it to the world soon. I got an award for creative writing, delivered to me by gunter globel, the nobel laureate, so that should help in passing the message. The portuguese ministery of education is doing a good job, in a bad way. The deal is to keep the evaluation as an exam, not by pairs otherwise there will be school police and even more politics and tension on the professors. Everyone should be evaluated and also the ministery. I think the most important aspect of it all is a question still unsolved, should we force people to study? How about giving the kids responsibility since they are young (13,14 y old) to decide wether or not to progress to more technical studies or academia. How to develop a taste for reading and curiosity, and to trigger creative minds is certainly a problem for days to come. I will do my part in advising my students and talking with my colleagues about this. I am starting to develop a taste for teaching, as I did not realize how the world needs people that know how to communicate and inflammate young hearts.

Imagine a place where money is not a problem. Imagine a place where people, smart and dum, because variety is essential, think together, work together. One week of reading and thinking, with 30 min of daily exercise to keep the body in shape. Imagine a place where these students are giving the freedom to create without absence of internal pressure, just inside competition. Imagine a place where art meets science, free from politics and from senile regulations. Imagine a set of competent rebels advised by experienced professionals from other fields outside science. This is evolution, using the technology of the XXIst century and the free thinking of the ancient socratic ages. This will be my institute of choice, now and for ever. For that I need to create a strong basis of support. And it will be seeded in Mirandela, after I build an airport there. We have to unleash creative minds into the world. The students will stay there for max duration of 4 years, where they will undergo extensive training and discussions. The cheapest reagents and organisms will be available and when required, the students will be sent to world wide conferences and institutes to perform their technical experimental requirements. THis is my Vision. It was taught to me by Iva, Kai and Carsten. We need a tough guy, to discipline rebel minds, so Wolfgang, Detlev and Joao Matos will be needed. We need motivated people, like Miguel, Monica and Iva. We need Kai because he is pure. We need Carsten because he has vision. Lets balance things, lets make good science happen in poor countries and change the system. Equilibrium, this is my destiny. I feel it, I know it, I will work for it. No hierarquies, no politics, no depression, euphoria on a leash and a big dose of self confidence and respect. We can do it, lets do it together.

Meus amigos. YES WE CAN. We can get an african american whose grandmother passed away recently as the president of the USA. He has no connections to the slaves lineages of the past, and he smoked weed when he was a kid. The man knows. But keep in mind, the NRA sold the double when he was elected. I just met some army troopers from California, and they were extremely concerned with the safety of their country, now that obama is in the White house. So lets say the man is the next kennedy, lets say the NRA didnt unleash Sarah Palin on purpose to loose the elections and increase the internal revenue of weapons. Because USA is a war machine, war outside, international pressure, fear, war inside... cool down period then the cycle begins.
China is making an airplane carrier, russia will become medieval due to corruption, and with Korea no one messes. Behind the curtains, Europe is a Switzerland, China is happy as long as the dollar is lower than the euro. Korea and India accept the investment from the multinational companies in the states.

As long as the economy holds we will not have a 3rd world war. Wath I predict is that after the states run out of African and eastern countries to exploit, europe should be prepared. We need a strong european union, with a central office of novelty, because the only way we can survive is to create inventions that no other country has, and wishes for. Thats the only way to export. Pos doc in the states? Maybe later when things calm down. Now I do not trust the extra couple of millions riffles out there.

Science, focus on it. We are pure and humble, and we wish to know and build knowledge, not to destroy it.

The most valuable lessons I have learned:
- You cannot save everyone, especially the ones who do not want to be saved.
- Focus on your own scientific problems, let other things be a side hobby
- Be organized, it saves time and you feel better when you come to work
- Say things only once, speak when you are sure about things. People will listen
- Read before you go to people. The you can get much more out of discussions.
- Communicate, do not be afraid - there are no things as ranks or patents in science.
- Listen carefully when others are speaking, do not discard anyone at anytime.
- Do one thing at a time. Plan, execute, analyze, put hypothesis, think about the outcome. Then re-start the cycle.
- Everyone gets depressed during their Phd, it is part of it. Rise up, stand up, learn the value of rest and enjoy the moments off the lab. Do not live in the institute, but work on it, use it as a tool for knowledge.
- How to say NO to people, It hurts, but it gives you freedom. And say what the fuck, it gives you even more freedom.
- The worst foes can become the best alies when you create opportunity.
- Patience, the basis of it all, a Phd is about learning how to manage your time, think about science, and patience, lots of it.
- Live, explore, travel. As Iva said "You are free persons, you are extremely talented and bright and you owe it to yourself to work hard, not to me, not to anyone, just to yourself!"
- Try again, think OUTSIDE THE BOX - this I knew before, now I have to think inside
it.
- Take your time to perform the experiments, get to breathe in between, do it stepwise, as kate said "Step by step procedure".
- Maté, Nikolai, Claudio, Matt, Catherine, Ana, Marina, Dina, Nicola,Maria, Marcus, Xico,Iana, Isabel, Phedon, João and Violeta, everyone in my city is amazing, and the sun is shining and the weather is sweet when these guys are around.
- Laughing, alongside with tenderness and relaxation,is indeed, without any doubt, the best thing in the world.
- Play, without hurting others, do exercise, get to know your physical limits. Explore, invent, and above all have fun. Life is measured by the impact you have on other lifes, let it be intense, smooth or just a glimpse. Enjoy!

February - One month non-stop action

The new students that applied for the MPI-CBG were welcomed by the 1st year Predocs (us) with a tour to the night-life in Dresden, a show, a movie and a party! From filling balloons, to impersonating half-naked neanderthals inside IKEA, I did a little bit of everything to make this big project more colourful! We were actors, stuntmen, dancers, talk-show hosts, directors and cameramen for two months, every weekend. And we did it, while working in the lab. Not so much sleep, skipped a few meals, but I am alive. Just arrived from Berlin, a fantastic conference on Microlfuidics and Biological separations, with Staffan Nillson, the man who levitates cells acousticaly and analysis secretion through mass spectrometry- And yeah, the water in the drop does not evaporate, because you can shoot more water in! :)

Ok my friends, here was the reason for my non-existing posts: Cold, study, TAC (Thesis Advisory Committee) and Ininterrupt Long term time Lapse microscopy. And acting, filming, directing and van driving a couch around Dresden. Yes, all for the Phd selection week partY!

The return of the gastarbeiter happened in the start of January.

The focused student, read almost everything there is about aging, wrote his Initial report and presented the project.
The aging field is a mixture of organism specific observations, centered around genomic instability, telomeres and mitochondria, and more discussion than actual experimental data. At least I have a lot to find out, the numbers, where are they? How many cell divisions can a cell undergo after all?

Some nice perspectives, also future uncertainties, proned him into deep introspection. The microfluidic chamber was a disappointment, no focus there, and back to the good old agarose patches. But he is stuborn, so now a new microfluidic setup will be born, from micro-laser litography and epoxi glue sensitive to UV.

The battle continues, between the bad (me), the good (the cells) and the vilan (the focus on the microscope). The cell tracking program is almost finished, still it requires some fine tuning.

Focus on immortality

It finally struck me. It came with a feeling of sadness and understanding. Like the scene of filming a paper bag in "American beauty". It is great to have ideas, but you can be successful with just hard work. Pick an idea and follow it, take it to the extremes of knowledge and logic. When you understand what you find then, open the chest, take another idea. Step by step. Logically, thoroughly, systematically unraveling life's little secrets.

That is true. I do not drink anymore. I refuse to ingest beer, wine is fine during dinner. But inside the clubs, if you want to find me, look for the guy holding the coke bottle! This way, I can go out on Fridays and Saturdays. I am eager for the smoking ban in January. Then going out is just like going to the gym, dancing and exercizing!

Washroom. Imagine a place where all the girls are beautiful. And nice. The music is divine, the dj is pumping. Now imagine meeting them all, and the cool guy friends,
Dancing forever in a stage in the middle of the floor, sharing drinks,
photos, phone numbers and messages. Now put Claudio, Radiosa and Analissa
in the picture. And the lights, the dancers, the artificial snow.

The cherry on top of the cake: My hat went around every head in the
club. I met 50 different people i dont remember half of the names. All good
looking, coming to me and smiling, And grooving, and smelling well. Sharing drinks, chanting PORTUGAL PORTUGAL! and huging getting me inside their circle of friends. I met everyone there. At some point I found myself wearing a shirt around my neck as an echarpe, then holding those sparkly candles with my teeth and dancing. Magic moment forever lost in time and space, but kept inside, really deep.

Dresden is the place to be, DEFINITELY. WITHOUT ANY DOUBT like Coutinho said.

Guess where we are going next saturday, to meet these guys in the next
major party. Washroom rocked, and I was alpha and omega. Dresden is my
city. Here i can be wathever i desire. Here I am at my full potential.

When you find something that you do not expect to find, you have a feeling of surprise. If it is an idea, you feel illuminated. It is like going to a street market in Morocco, walking the streets and discovering new things in every tent. New smells, new lights, pretty little treasures just around the corner. The diversity of life can only be compared to the grandure of feelings. Love, passion, hate, Joy, anger. It is all a matter of contrast as we are made by machines that measure potentials and adjust to them. Adaptation and Evolution, Yien and Yang. Benfica and Porto. Science and Religion. We need them all, as without one the other would be meaningless. I wonder if during evolution we will discover new feelings. Can you imagine a colour that does not exist? It is like reaching to the spiritual world. The colour that does not exist can find you first.

Tomorrow I will go to the Nanodogs and Nanocities - 1st part sponsored by the DIGGS-BB fellowship I got through the Phd office. The leading edge science on micro-machines and nanostructures will be presented in two intense days at the Steigenberger Hotel, in front of the Frauenkurche (Our Lady's Church). I cannot sleep, the heart runs fast, the ideas flow and the world starts to feel small. Focus, Kai said, and that is what I will do in the next few days. Kai Simons, pretty intense discussion about protein targets for the transition state analogs. Two candidates- Thrombin and Transferrin. Immune system tests with enzyme bound to hapten injected into rabbits. Great idea! Focused and ready, here I am reborn.

In the afternoon we looked at the regenerated axolotl tails, to see whether the overexpression of target genes would have an impact on Pax7 levels. Indeed one of the most promising single results I had! Thanks Akira, for the lectures and the kindness.

After going to the 1st Italian restaurant in Alaunstrasse, had a Pomodoro soup, a Prosciutto Pizza and for dessert Spaghetti Salmone! Whonzimmer until 12h, then we had a n invitation from Dj Ulf, that Nikolai met at Korrale, for a party in Industrigeland. We we got there, we entered for free, me Nikolai, Pear and Jen. Little we knew that Ulf was the best dj in Dresden, in the easy going way that he showed towards us. Dresden is full of surprises! Amazing party with an extra, the One Ampere rock band that filled the house, beauty all around, and Ellen Alien, Knife remixed and Michael Andrews on the dancefloor. Donnie Darko had quite a night my friends. This one will stay forever, stopped in time, like tears in rain.

Dresden, I love you! Just had a warm bubble bath and going to bed, not before I recorded every single moment. And yes, I had all the fun a guy can get when drinking coca-cola all night long! Jane, you should come next time, definitely!

Regeneration. What if you could grow back our limbs like these little Axolotls that are able to regenerate virtually any body tissue and nervous system. Funny little creatures that enclose one of the secrets to the fountain of youth? We will study gene expression in the regenerated tail. Injection in the spinal cord is a laborous process, but when one gets the hang of it, GFP flows through, and after electroporation shines inside the nervous cells. The poor guy, G2 piercing, had a 4mm capilar tip broken roaming inside the notocord. Akira saved him with the scalpel and he now has a "bright" future ahead. Hail to the lizard king, the Axolotl. :) One curiosity: These animals are neotenic, which means they are arrested in development. Nevertheless, proliferation can be distinguished from de-differentiation using non-proliferative muscle cells.

The meeting with Dr Werner came early in one of the best Fridays in my life! I had time to prepare the project with everything I learned this week in the Werner lab, about biomaterials, physics, and blood interactions. I am in debt to Mirko, Martin, Phillip, Uwe, and to the rest of the guys in the lab who made this week feel like heaven. The best practical course for sure, well organized and professional. Evolution. At least in the physics field, I grew as a scientist. Incredibly well driven hands-on week-course with single DNA molecule dynamics analysis. For a biochemist it was beautiful to see a single Type I endonuclease (a protein that cuts the DNA as serves as a defense mechanism in bacteria when they are infected with viral DNA) looping the DNA and then cooperating to cut it!) cleaving a DNA molecule live! Real time analysis of single molecule events, taking one step further into the unknown.

The bionic human. Who doesn't remember the androids from blade runner, facing the problem of limited lifespan and loving life more than anything in the end. Nowadays we face the same problem: what separates the use of artificial parts inside human beings is the resilience to the attack of the Immune System. Yesterday lecture from Werner combined with my hyper-threading synapses after an entire week of brainstorming may lead to the fusion of science and science fiction. I came to cross several fields and developed a clever way of bypassing the immune attack. In the end it might really work. I could not sleep for too long. I feel the rush of ideas and now I have to shape them into projects. Possibly another collaboration, with a major impact in industry, medicine and the human health. Another step towards the fountain of youth? I feel alive, I feel well. Sometimes is as if I could see into the future. Now I just have to paint it!

Dresden, das angebote. Finally here, my friend the polish-american-uruguayan-brazilien-portuguese Patryk. The fotos speak for themselves, and anyway I am too tired after waking up at 5am today, so my poetry will not be used today. We had loads of fun, it started whit the party and with the ALOOOOOOOO girl behind me, and continued with J-Lo and the Queen of Finland as our special guests. But the main event was when Patrik went to Jean Leon's room, and this amazingly beautiful Arabian girl opened the door and gazed at him surprised when he asked "Did you see Miguel, he disapeared yesterday and I am looking for him!" He held his temper,and acted cool, just like Guy Cooney, when interviewed by the BBC! So in the end, the prank was even more funny when another girl that was at my party appeared, and told him "Why dont you just call him, you have his phone number!".

We ended up meeting lots of new people. I can only say, that the octopus guy from resident evil is the king of disco, with all those strobs decorating the place, and that mila jovovich is still Mila. Gotta go now, nap time, and a well deserved one I might say. Near Field microscopy RULEZZZZZZZ!

Its been 50 days in Germany. Nice results, flat finally assembled, cooking style flowing, and wireless internet starting today! Working on weekends and ikeaing with multiple van types and configurations. Black & Decker almost done. Now I just need to put up the curtains and prepare myself for the lectures. Science, just another link, just another question, one more experiment.
